目的 建立基于超高效液相色谱串联高分辨质谱法测定谷氨酸钠注射液中谷氨酸钠含量的高灵敏和高选择性新方法。方法 色谱柱为Kinetex C18(2.0×100mm,1.7μm),流动相为0.1%甲酸水和甲醇溶液,样本中谷氨酸以3-溴丙基三苯基磷(3-BMP)进行衍生化反应,流速为0.40μL/min,谷氨酸衍生化物母离子和特征碎片分别为450.18→320.16。使用非数据依赖型扫描模式绘制标准曲线。结果 谷氨酸钠在0.024~1000pmol范围线性关系良好(R2=0.9999),最低检出量为4.00fmol,最低定量限为24.25fmol,平均加标回收率为91.71%,日内日间精密度RSD分别为1.67%~2.64%和1.66%~4.55%。结论 该方法简便、灵敏、准确,样品处理简单,适用于产品质量检测和实验室中测定谷氨酸钠注射液中谷氨酸钠的含量。 |

Objective To establish an ultra performance liquid chromatography tandem high resolution mass spectrometry for the determination of sodium glutamate in sodium glutamate injection.Methods Kinetex C18 (2.0×100mm,1.7μm) was used as the chromatographic column,and the mobile phase was 0.1% formic acid water and methanol solution.In the sample,glutamic acid was derivatized with 3-bromopropyl triphenylphosphorus (3-BMP) at a flow rate of 0.40L/min,and the parent ions and characteristic fragments of glutamic acid derivatives were 450.18→320.16,respectively.Draw standard curves using data-independent scanning mode.Results The linear relationship of sodium glutamate was good in the range of 0.024~1000pmol (R2=0.9999),the minimum detection limit was 4.00fmol,the minimum quantitative limit was 24.25fmol,the average recovery rate was 91.71%,and the RSD of intra-day and inter-day precision were 1.67%~2.64% and 1.66%~4.55% respectively.Conclusion The method is simple,sensitive and accurate,and the sample processing is simple.It is suitable for product quality inspection and laboratory determination of sodium glutamate in sodium glutamate injection. |
