      目的 基于GDC TCGA Breast Cancer (BRCA)的数据,评估TRIM45(Tripartite Motif Family 45)在乳腺癌中的表达、预后价值及其与临床病理的相关性,揭示TRIM45在乳腺癌中的生物学功能及可能的作用机制。 方法 采用R软件对GDC TCGA Breast Cancer (BRCA)数据进行生信分析,研究TRIM45在乳腺癌中的表达及其与临床病理的关系。采用Kaplan-Meier法评估TRIM45对乳腺癌预后的影响。应用KEGG基因集进行GSEA(Gene Set Enrichment Analysis)基因富集分析,采用Spearman相关性分析筛选TRIM45相关作用基因。 结果 TRIM45在乳腺癌临床样本中的表达高于癌旁组织(P<0.05);Kaplan-Meier生存分析显示TRIM45高表达患者的Overall Survival、Disease Specific Survival和Progression Free Interval均高于TRIM45低表达者(P<0.05);TRIM45在乳腺癌中的表达与T分期、TNM分期、分子分型及病理类型相关,并且随着乳腺癌恶性程度的升高,TRIM45的表达在T分期、N分期、M分期和TNM分期中的表达逐级下降;GSEA富集分析结果显示TRIM45在KEGG_PATHWAYS_IN_CANCER信号通路中发挥重要的生物学作用,Spearman相关性分析筛选出TRIM45与STK36、IKBKB、GLI3具有正相关关系,与HIF1A、ETS1具有负相关关系。 结论 TRIM45在乳腺癌中的表达与T分期、TNM分期、分子分型及病理类型相关,且随着乳腺癌恶性程度的升高,TRIM45的表达逐级下降;TRIM45在乳腺癌中为抑癌基因,可能通过调控STK36、IKBKB、GLI3、HIF1A和ETS1在乳腺癌发生发展中发挥重要作用。

      Objective Based on the data of GDC TCGA Breast Cancer (BRCA), to evaluate the expression, prognostic value and clinical pathology correlation of TRIM45 in breast cancer, revealing the biological function of TRIM45 in breast cancer and its possible Mechanism. Method Bioinformatics analysis of GDC TCGA Breast Cancer (BRCA) data was performed using R software to study the expression of TRIM45 in breast cancer and its correlation with clinical pathology; the Kaplan-Meier method was used to evaluate the effect of TRIM45 on the prognosis of breast cancer; the KEGG gene set was used for GSEA gene enrichment analysis; Spearman correlation analysis was used to screen TRIM45-related genes. Results The expression of TRIM45 in breast cancer was higher than that in adjacent tissues (P<0.05); Kaplan-Meier survival analysis showed that the Overall Survival, Disease Specific Survival and Progression Free Interval of patients with high TRIM45 expression were higher than those of patients with low TRIM45 expression (P<0.05); the expression of TRIM45 in breast cancer is related to T stage, TNM stage, molecular type and pathological type; and with the increase of the malignancy of breast cancer, the expression of TRIM45 gradually decreased in T, N, M and TNM stages; GSEA enrichment analysis showed that TRIM45 plays an important biological role in the KEGG_PATHWAYS_IN_CANCER signaling pathway; Spearman correlation analysis screened out that TRIM45 was positively correlated with STK36, IKBKB and GLI3, and negatively correlated with HIF1A and ETS1. Conclusion The expression of TRIM45 in breast cancer is related to T stage, TNM stage, molecular typing and pathological type. With the increase of the malignant degree of breast cancer, the expression of TRIM45 gradually decreased in T, N, M and TNM stages. TRIM45 is a tumor suppressor gene in breast cancer. It may play an important role in the occurrence and development of breast cancer by regulating STK36, IKBKB, GLI3, HIF1A and ETS1.
