      摘 要 目的 探讨电针对缺血性脑损伤后GRP78和caspase12的影响及对大鼠肢体运动功能修复的影响及机制。方法 本实验使用改良线栓法制备大鼠脑缺血再灌注模型(MCAO)[1],对“百会”和“水沟”穴电针刺激,电针干预时间30 min。随机将65只SD大鼠分为对照组(sham)、脑缺血再灌注3 h、6 h、12 h、24 h、72 h、120 h组(I/R)和再灌注时间点+电针组(I/R+EA),n=5,用于RT-PCR检测caspase-12 mRNA的表达及检测GRP78蛋白的表达;35只SD大鼠脑缺血再灌注术后及电针干预7 d后,于再灌注7 d、14 d和30 d分别做大鼠转棒式疲劳仪实验,检测大鼠肢体运动功能的修复状况。结果 电针能下调大鼠缺血侧脑组织caspase12 mRNA的表达; I/R+EA组再灌注3 h、6 h、12 h 的GRP78阳性细胞数与对应时间的I/R组相比明显减少(P<0.05);大鼠转棒式疲劳仪实验结果显示电针干预能促进大鼠肢体运动功能的恢复(P<0.05)。结论 电针干预能促进缺血性脑损伤大鼠肢体运动功能修复可能与阻断内质网介导的凋亡通路有关。

      Abstract:Objective To investigate the effect and possible mechanism of electroacupuncture on cerebral cell apoptosis and neural function repair in rats with ischemic stroke. Method Simple random sampling will divide 65 rats into control group (sham), groups of cerebral ischemia reperfusion for 3 h, 6 h, 12 h,24 h,72 h,120 h(I/R) and groups of cerebral ischemia reperfusion for 3 h, 6 h, 12 h,24 h,72 h,120 h and acupuncture intervention (I/R + EA). Each group has 5 rats. Use thread bolt method to make model of cerebral ischemia reperfusion in rats(MCAO). Stimulate on "baihui" and "shuigou" acupoint. The intervention time of electroacupuncture was 30 min. RT-PCR detected the expression of caspase12 mRNA in the cerebral cortex and striatum of rats,fluorescent histochemical method detected the expression of GRP78 protein.After cerebral ischemia and reperfusion for 35 SD rats and 7 days after the intervention with electroacupuncture, rotator fatigue tests were performed on 7 d, 14 d and 30 d after reperfusion to observe the repair of body movement function in rats. Result RT-PCR results showed that the expression of caspase12 mRNA was reduced after electroacupuncture; The immunofluorescence histochemical results showed that the number of GRP78 protein-positive cells and the corresponding I/R group were significantly reduced after electroacupuncture intervention after reinfusion of 3 h, 6 h, and 12 h(P<0.05);The experimental results of the rod-type fatigue instrument in rats showed that the motor function of the rats improved obviously after electroacupuncture.Conclusion Acupuncture intervention may prevent brain cell apoptosis and promote the repair of body movement function after cerebral ischemia-reperfusion injury by blocking apoptosis pathway mediated by endoplasmic reticulum and inflammatory factor.
