      目的 制备牛血清白蛋白(BSA)温敏凝胶给药系统,并考察给药系统的释放情况。方法 以温敏凝胶材料普朗尼克F127为载体,以相变温度为指标,通过倒置试管法筛选普朗尼克F127的浓度。通过BCA蛋白定量试剂盒法测定BSA的含量。以PBS缓冲液为释放介质,考察凝胶给药系统中BSA的释放。结果 浓度为20%(w/v)的普朗尼克F127凝胶的相变温度为23℃,载入BSA后相变温度不变。BSA在普朗尼克F127凝胶系统中的释放12h可释放80%以上,具有缓释效果,且释放接近零级动力学过程。结论 BSA-普朗尼克F127温敏凝胶系统制备方法简单,具有明显的缓释效果。

      Objective To study on the preparation of bovine serum albumin (BSA) loaded temperature-sensitive gel and the release property of BSA. Methods Pluoronic F127 was choosed as carriers. The concentration of pluoronic F127 was optimized through inverted test tube method using phase-transition temperature as index. The concentration of BSA was determined using BCA kits. The release property of BSA was investigated using PBS as dissolution media. Results The phase-transition temperature of pluoronic F127 with the concentration of 20%(w/v) was 23℃ and retained the same when BSA was loaded. The release results showed that after 12h, more than 80% of BSA was released and the release property was nearly zero-order kinetics. Conclusion The preparation technique of BSA loaded pluoronic F127 temperature-sensitive gel is feasible. The release of BSA could be well controlled.
