      目的 基于16s rRNA测序分析微小RNA-625-3p(miR-625-3p)基因沉默对结直肠癌小鼠的肿瘤抑制作用及对肠道菌群的影响。方法 制备、获取miR-625-3p基因沉默(Si)和沉默阴性对照(SiNe)外泌体。BALB/C小鼠腹腔注射SW620细胞(5×106),随机分为模型(A)组,奥沙利铂(Oxaliplatin,B)组,Oxaliplatin+exo-miR-625-3p-Si(C)组和Oxaliplatin+exo-miR-625-3p-SiNe(D)组,每组3只。观察miR-625-3p基因沉默对结直肠癌小鼠肿瘤体积、肿瘤细胞凋亡、肠道内容物菌群丰度及多样性的影响。结果 成功分离出miR-625-3p-Si和miR-625-3p-SiNe外泌体并鉴定。外泌体干预后,与D组比较,C组miR-625-3p表达降低(t=-4.057,P<0.05),肿瘤体积减小(t=-2.331,P=0.08),凋亡增加(t=2.945,P<0.05);OTUs数量降低;门水平上,Bacteroidetes丰度升高(t=3.590,P<0.05),Firmicutes丰度降低(t=-3.608,P<0.05);筛选出C、D两组间存在显著差异菌种29种。结论 MiR-625-3p基因沉默能抑制结直肠癌的发展,促进结直肠癌细胞凋亡,其机制可能与改善小鼠肠道微生态环境有关。

      Objective The effects of microRNA-625-3p (miR-625-3p) gene silencing on tumor inhibition and intestinal flora in colorectal cancer mice were analyzed based on 16s rRNA sequencing.Methods Prepare and acquire of miR-625-3p gene silencing (Si) and silencing negative control (SiNe) exosomes.SW620 (5×106) was intraperitoneal injected and divided into model (A) group,Oxaliplatin (B) group,Oxaliplatin+exo-miR-625-3p-Si (C) group and Oxaliplatin+exo-miR-625-3p-SiNe (D) group,three mice per group.To observe the effects of miR-625-3p gene silencing on tumor volume,tumor cell apoptosis,abundance and diversity of intestinal flora in colorectal cancer mice.Results Exo-miR-625-3p-Si and exo-miR-625-3p-SiNe were successfully isolated and identified.After exosome intervention,compared with group D,the expression of miR-625-3p in group C decreased (t=-4.057,P<0.05),tumor volume decreased significantly(t=-2.331,P=0.08),and apoptosis increased(t=2.945,P<0.05).The number of OTUs decreases;at the phylum level,the abundance of Bacteroidetes increased (t=3.590,P<0.05)and the abundance of Firmicutes decreased (t=-3.608,P<0.05);Twenty-nine strains with significant differences between the C and D groups were screened.Conclusion MiR-625-3p gene silencing can inhibit the development of colorectal cancer and promote the apoptosis of colorectal cancer cells,which may be related to the improvement of the intestinal microecological environment of mice.
