      目的 合成海藻酸钙/透明质酸/羟丙基-β-环糊精复合水凝胶,研究透明质酸对该凝胶的影响。方法 利用溶胶-凝胶法合成该复合凝胶;使用光学显微镜(OM)、扫描电子显微镜(SEM)、傅里叶红外光谱(IR)、X射线衍射(XRD)、流变学等研究凝胶结构、性质性能等;采用牛津杯法测试左氧氟沙星凝胶制剂的抗菌效果。结果 透明质酸以氢键结合海藻酸钙,妨碍了海藻酸钙凝胶的多糖链带状和网状微结构的聚集,增大了网状空隙而利于载药;其左氧氟沙星凝胶制剂作用于金黄色葡萄球菌和大肠埃希氏菌,抑菌效能可分别达到对照标准的85.2%和91.7%。结论 透明质酸优化了海藻酸钙复合凝胶的微结构和性能;新型复合凝胶合成方法简便,其左氧氟沙星凝胶制剂具有良好的抑菌效果。

      Objective A composite hydrogel based on calcium alginate (CA) with hydroxypropyl β-cyclodextrin (HPCD) is synthesized in presence of hyaluronic acid (HA), and its micro-structure, physical properties and formation mechanism are investigated.The antibacterial effect of levofloxacin (LVFX) loaded within hydrogel will be verified.Methods The composite hydrogel was synthesized by sol-gel method, and its structure and properties were studied by optical microscopy (OM), scanning electron microscope (SEM), Fourier infrared spectroscopy (IR), X-ray diffraction (XRD) and rheology.The antibacterial effect of the gel with levofloxacin was tested by Oxford cup method.Results The composite hydrogel was successfully acquired.The molecular chains of HA entangled with the molecular straps of CA relying on H-bond force, which hindered CA chain aggregation or expansion of CA gel network.This resulted to more holes or interspace within the hydrogel network for drug loading.The antibacterial efficacy of LVFX within the hydrogel could reach about 85.2% and 91.7% to the positive antibacterial control on Staphylococcus aureus and Escherichia coli, respectively.Conclusion The composite gel can be acquired in a direct and easy method.The microstructure and properties of CA/HPCD hydrogel were optimized by the participation of HA.The new composite hydrogel deserves to be widely applied as drug carrier.
