| 目的 探讨长链非编码RNA MIR4435-2HG与miR-128-3p在膀胱尿路上皮癌中的表达意义及相关性。方法 采用实时荧光定量PCR检测87例膀胱尿路上皮癌组织及癌旁正常组织MIR4435-2HG和miR-128-3p的表达，将患者分别分为MIR4435-2HG高表达组（44例）、MIR4435-2HG低表达组（43例）或miR-128-3p高表达组（39例）、miR-128-3p低表达组（48例）；比较两组间病理资料和预后生存差异；分析MIR4435-2HG和miR-128-3p表达的相关性。结果 膀胱尿路上皮癌组织中MIR4435-2HG表达水平明显高于癌旁组织，而miR-128-3p表达水平明显低于癌旁组织（P<0.05），且MIR4435-2HG和miR-128-3p表达呈负相关（r=-0.544，P<0.05）。MIR4435-2HG低表达和高表达组在病理分级、临床分期和淋巴结转移方面有统计学差异（P<0.05），而miR-128-3p低表达和高表达组各资料均无统计学差异（P>0.05）；MIR4435-2HG高表达患者生存率低于MIR4435-2HG低表达组，而miR-128-3p高表达患者总生存率高于miR-128-3p低表达组（P<0.05）。MIR4435-2HG高表达和miR-128-3p低表达是影响膀胱尿路上皮癌患者预后的独立危险因素。结论 膀胱尿路上皮癌组织MIR4435-2HG表达上调，miR-128-3p表达下调，两者均是患者预后不良的危险因素。 |

| Objective To investigate the expression significance and correlation of long non-coding RNA MIR4435-2Hg and miR-128-3p in urothelial carcinoma.Methods The expressions of MIR4435-2HG and miR-128-3p in 87 cases of bladder urothelial carcinoma tissue and adjacent normal tissues were detected by real-time quantitative PCR.The patients were divided into high MIR4435-2HG group (44 cases) and low MIR4435-2HG group (43 cases), high miR-128-3p group (39 cases) and low miR-128-3p group (48 cases);the difference of pathological data and prognosis between the two groups was compared, and the correlation between the expression of MIR4435-2HG and miR-128-3p was analyzed.Results The expression level of MIR4435-2HG in bladder urothelial carcinoma tissue was significantly higher than that in adjacent tissues, while the expression level of miR-128-3p was significantly lower than that in adjacent tissues (P<0.05), and the expression level of MIR4435-2HG and miR-128-3p was negatively correlated (r=-0.544, P<0.05).There were statistical differences in pathological grade, clinical stage and lymph node metastasis between the low and high MIR4435-2HG group (P<0.05), while there were no statistical differences in all data between the low and high miR-128-3p group(P<0.05);The survival rate of patients with high MIR4435-2HG was lower than that of low MIR4435-2HG group, while the overall survival rate of patients with high miR-128-3p was higher than that with low miR-128-3p group (P<0.05).High MIR4435-2HG and low miR-128-3p were independent risk factors for prognosis in patients with bladder urothelial carcinoma.Conclusion The expression of MIR4435-2HG in bladder urothelial carcinoma tissue was up-regulated, and the expression of miR-128-3p was down-regulated, both of which were risk factors for poor prognosis. |
