| 目的 探讨血清饥饿诱发的前列腺癌DU145细胞适应性反应。方法 血清培养DU145细胞24h后更换为无血清培养基。24、48、72h后收集细胞，分别应用MTT实验、细胞计数实验研究血清饥饿对细胞活力、增殖能力的影响；Real-time PCR和Western blot实验分别在mRNA水平和蛋白水平检测干预前后细胞上皮间质转化（Epithelial-Mesenchymal Transition，EMT）相关基因表达的变化。结果 随着血清饥饿时间的延长，DU145细胞拉长呈细叶状，多中心聚合性生长；细胞活力、增殖能力受到明显抑制。Real-time PCR、Western blot结果均显示血清饥饿后E-钙黏蛋白（E-cadherin）表达升高，SNAIL、SLUG表达降低。结论 为应对血清饥饿，DU145细胞发生了EMT的逆转，这可能是其适应性反应的一部分。 |

| To explore the cellular adaptive response of human prostate cancer DU145 cells induced by serum starvation.Methods Prostate cancer DU145 cells were cultured in DMEM supplemented in 10% FBS and then replaced with serum free medium.Cells were collected from serum and serum-free medium after 24,48 and 72 hours.The cell viability and multiplication capacity were detected using MTT and Cell Counting,respectively.RT-PCR and Western Blotting are used to examine EMT related gene expression.Results With serum starvation morphological changes of DU145 cells were observed.MTT and Cell counting results indicated that serum starvation inhibited cell proliferation significantly.Real-time PCR and Western Blotting results showed that the expression of E-cadherin gene increased,while SNAIL,SLUG decreased.Conclusion In response to serum starvation,EMT reversion occurred in DU145 cells and this performance may be a part of the adaptive response. |
