| 目的 观察雷公藤红素对神经母细胞瘤SK-N-SH细胞增殖的抑制作用,并探讨其作用机制。方法 培养SK-N-SH神经母细胞瘤细胞株。采用CCK8法和克隆形成实验检测SK-N-SH细胞的增殖率,应用流式细胞仪分析细胞0 h、24 h、48 h、72 h细胞周期的变化情况,应用Western Blot检测SK-N-SH细胞中细胞周期相关蛋白Cyclin D1、P53、P21、GAPDH的表达,利用生物信息学分析雷公藤红素对神经母细胞瘤可能的作用靶点及作用机制。结果 雷公藤红素对SK-N-SH细胞的增殖有明显的抑制作用(P<0.01);流式细胞仪显示24 h、48 h、72 h的G0/G1期细胞持续增多;Western blot显示雷公藤红素下调CyclinD1表达,生物信息学分析显示雷公藤红素诱导细胞发生一系列反应,集中于细胞应激方面。结论 雷公藤红素显著抑制SK-N-SH细胞增殖,其机制可能是通过下调CyclinD1蛋白表达而发挥作用。 |

| Objective To investigate the effect of celastrol on proliferation of neuroblastoma SK-N-SH cells and the possible mechanism. Methods We treated cultured SK-N-SH cells with celastrol and CCK8 assay, colony formation assay was used to detect the proliferation of cells. The changes of cell cycle were assessed by flow cytometry. We used Western blot to measure the expression of CyclinD1, P53, P21, GAPDH, analyzing of possible targets and mechanisms of action of celastrol on neuroblastoma using bioinformatics. Results Celastrol significantly inhibited the proliferation of SK-N-SH cells in time-dependent manner (P<0.01). Flow cytometry showed that cells continued to increase on G0/G1 phase at 24 h, 48 h and 72 h. Moreover, the protein expression of CyclinD1 was markedly suppressed by celatrol. Bioinformatics analysis reveals that tretinoin induces a series of cellular responses focused on cellular stress. Conclusion Celastrol can inhibit the proliferation of SK-N-SH cells, probably through the down-regulation of expression of CyclinD1 protein. |
