      目的 观察桑寄生水提物对慢性不可预知温和刺激(chronic unpredictable mild stress,CUMS)模型小鼠抑郁样行为的作用及对脑中5-HT含量的影响。方法 SPF小鼠40只,随机均分为空白组、模型组、阳性药组、桑寄生水提物低剂量组和桑寄生水提物高剂量组,除空白组外其余组小鼠通过CUMS结合单笼孤养干预45 d,造模期间定时灌胃,阳性药组给予氟西汀0.006 g·kg-1·d-1,桑寄生水提物高和低剂量组分别给予桑寄生水提物2.5 g·kg-1·d-1、1.25 g·kg-1·d-1。在造模第1、9、18、27、36天称量小鼠体重并进行糖水偏好实验;造模第36天开始行为学测试包括旷场实验、强迫游泳实验和悬尾实验。第45天称量小鼠体重后断头取脑,ELISA检测小鼠大脑组织匀浆上清液中5-HT含量。结果 与空白组对比,模型组小鼠第27天起体重增长率明显降低(P<0.01);旷场实验中穿越中央次数显著减少(P<0.01),直立次数显著减少(P<0.01);强迫游泳持续不动时间显著增加(P<0.01);悬尾持续不动时间明显增加(P<0.05);小鼠脑5-HT含量显著降低(P<0.01)。与模型组比较,阳性药组第27天后小鼠体重均明显增加(P<0.01),在第27和45天,桑寄生水提物低剂量组和高剂量组小鼠体重增加(P<0.05);糖水偏好率变化均无统计学差异(P>0.05);旷场实验中阳性药组、桑寄生高剂量组小鼠穿越中央次数和直立次数显著增加(P<0.01);强迫游泳实验中阳性药组、桑寄生水提物高剂量组持续不动时间显著减少(P<0.01);阳性药组、桑寄生水提物低剂量组小鼠悬尾持续不动时间明显减少(P<0.05);阳性药组、桑寄生水提物低剂量和高剂量组小鼠5-HT含量均显著升高(P<0.01)。结论 桑寄生水提物对CUMS模型小鼠抑郁样行为有改善作用,其机制可能与增加脑组织中5-HT含量有关。

      Objective To observe the effects of taxillus chinensis water extract on depression-like behavior and brain 5-HT levels of chronic unpredictable mild stress (CUMS) model mice. Methods 40 SPF mice were randomly divided into blank group,model group,fluoxetine group,low dose and high dose taxillus chinensis water extract group (low dose group and high dose group).Except the blank group, mice in the other groups were treated with CUMS combined with single cage isolation for 45 days.During the modeling period, mice in fluoxetine group were given fluoxetine 0.006g·kg-1·d-1.The high-dose and low-dose groups were given 2.5g·kg-1·d-1 and 1.25g·kg-1·d-1,respectively.At the 1st,9th,18th,27th and 36th day of modeling,mice were weighed and sugar water preference experiment was carried out.Behavioral tests began on the 36th day of modeling,including open field experiment,forced swimming experiment and tail suspension experiment.On the 45th day,after final body weight measurement,mice were euthanized via decapitation and brains were extracted,and the content of 5-hydroxytryptamine (5-HT) in the supernatant of the homogenate of the mouse brain tissue was detected by ELISA. Results Compared with blank group,the growth rate of body weight in model group decreased significantly from day 27 (P<0.01).In open field experiment,the number of crossing the center was significantly decreased (P<0.01),and the number of upright was significantly decreased (P<0.01).The duration of forced swimming was significantly increased (P<0.01);The duration of suspension was significantly decreased (P<0.05);The content of 5-HT was significantly decreased by ELISA (P<0.01).Compared with model group,the body weight of mice in fluoxetine group at day 27 was significantly increased (P<0.01),and the body weight of mice in low-dose and high-dose groups was significantly increased (P<0.05).There were no significant differences in the sugar-water preference rate (P>0.05).In open field experiment, the crossing times and upright times of mice in fluoxetine group,high dose groups were significantly increased (P<0.01).The duration of immobility was significantly decreased in fluoxetine group,high dose groups (P<0.01). The duration of tail suspension in fluoxetine group,low-dose group were significantly decreased (P<0.05).The content of 5-HT in fluoxetine group,low dose and high dose groups was significantly increased (P<0.01). Conclusion The aqueous extract of Taxillus chinensis has an ameliorative effect on depressive-like behaviors in CUMS model mice,and the mechanism may be related to the increase of 5-HT content in brain tissue.
