      目的 建立高效液相色谱法-二极管阵列检测器(HPLC-DAD)法评价青钱茶对α-葡萄糖苷酶抑制活性的方法。方法 依据α-葡萄糖苷酶抑制剂可抑制α-葡萄糖苷酶对4-硝苯基-α-D-吡喃葡萄糖苷酶解的原理,以乙腈-0.3%乙酸为流动相,梯度洗脱测定4-硝苯基-α-D-吡喃葡萄糖苷酶解产物对硝基酚,检测波长315nm,流速1.0ml·min-1,柱温35℃。结果 对硝基酚在17.42~348.4μg·ml-1范围内线性关系良好,回收率为98.8%。结论 所建立的方法测定结果准确,重现性好,可用于评价青钱茶对α-葡萄糖苷酶的抑制活性。
英文摘要:
      Objective To establish an HPLC-DAD method for evaluation the activity of α-glucosinase inhibitors of Qingqian tea.Methods According to the principle that α-glucosidase inhibitors can inhibit the enzymatic hydrolysis of α-glucosidase to 4-nitrophenyl-α-D-pyranoglucoside,an HPLC method was developed to determine p-nitrophenol,the deglycoside product of 4-nitrophenyl-α-D-glucopyranoside,using gradient elution with acetonitrile-0.3% acetic acid as mobile phase.The flow rate was 1.0 ml·min-1 and the detection wavelength and column temperature were set as 315 nm and 35℃,respectively.Results P-nitrophenol was linear in the range of 17.42~348.4μg·ml-1.The average recovery was 98.8%,with RSD of 1.5%.The method was accurate and reproducible.Conclusion The method is sensitive and accurate, and can be used to evaluate the inhibition activity of Qingqian tea on α-glucosinase.
