摘要 目的 观察布洛芬对小鼠运动神经元轴索生长的影响并探讨其可能的分子机制。方法 利用密度梯度离心法分离孕13.5天的C57BL/6小鼠胚胎脊髓前角中的运动神经元,加入400μmol/L的布洛芬作为刺激,以神经生长因子(NGF)(100ng/ml)作为阳性对照,培养48小时后以轴索生长测定观察布洛芬对运动神经元轴索生长的影响,收集不同处理的运动神经元测定Rho激酶(ROCK)活性,荧光定量PCR检测MAPK信号分子(ERK, P38, JNK)的表达。结果 布洛芬组神经元轴索长度为89.1±10.7μm,与空白组(66.5±8.4μm)相比有显著性差异(p?0.001),与NGF组(86.0±11.8μm)相比差异无统计学意义(p=0.732)。布洛芬组神经元中ROCK活性显著低于空白组和NGF组(均有p?0.001)。荧光定量PCR显示布洛芬组JNK mRNA的相对表达显著高于其余两组(均有p?0.001),三组间ERK和p38的表达均无显著性差异。结论 布洛芬可能通过上调JNK的表达而促进运动神经元轴索的生长。 |

Abstract:Objective To investigate the effects of ibuprofen on axonal growth of motor neuron and explore its possible mechanism. Methods The primary motor neuron was prepared from the embryonic spinal cord of C57Bl/6 mouse on day 13.5 using the density gradient centrifugation and cultured with different
treatment, blank, ibuprofen (400μmol/L) and nerve growth factor (NGF) (100ng/ml). After 48 hours, axonal growth assay was performed to investigate the effects of ibuprofen on axonal growth of motor neuron. The neurons with different treatment were collected for Rho kinase activity and Real-time PCR was performed to investigate the relative expression of MAPK signaling pathway molecules (ERK, P38, JNK). Results The axonal length of ibuprofen-treated motor neuron (89.1±10.7μm) was significantly longer than that of motor neuron without treatment (66.5±8.4μm) (p?0.001). There was no difference in axonal length between the ibuprofen-treated and NGF-treated motor neuron (86.0±11.8μm) (p=0.732). The Rho kinase activity in ibuprofen-treated motor neuron was significantly lower than that of both NGF-treated motor neuron and blank controls (p?0.001, both). The PCR analysis showed that there was higher relative expression of JNK mRNA in ibuprofen-treated motor neuron than the other two groups (p?0.001, both). There was no difference in relative expression of both ERK and p38 among the three groups. Conclusion Ibuprofen may promote the axonal growth of motor neuron by upregulating the expression of JNK and inhibiting the Rho kinase activity. |
