摘要 目的 初步探讨花生红衣水提物对小鼠的镇静催眠作用及作用机制。方法 采用小鼠自主活动测试仪观察记录小鼠自主活动,采用协同戊巴比妥钠致小鼠睡眠实验观察花生红衣提取物对小鼠的镇静催眠作用,酶联免疫吸附法(ELISA)测定小鼠脑组织中γ-氨基丁酸(GABA)含量。结果 低剂量组花红衣水提物可抑制小鼠自主活动能力(P<0.05),增加阈下剂量戊巴比妥钠小鼠的入睡只数(P<0.05),缩短阈上剂量戊巴比妥钠小鼠的入睡潜伏期(P<0.01),延长睡眠持续时间(P<0.05),升高小鼠脑组织中GABA含量(P<0.01)。结论 花生红衣水提物有一定的镇静催眠作用,其作用机制可能与提高小鼠脑内GABA水平有关。 |

Abstract: Objective To investigate the Sedative and hypnotic effects of peanut red skin extract and to explore the potential mechanism. Methods Sedative-hypnotic experiments were conducted to observe the spontaneous activity in mice and its sleep effect on the superthreshold and subthreshold doses of sodium pentobarbital. ELISA method was employed to detect the contents of GABA in brain of mice. Results Low doses of peanut red skin water extract significantly reduced the number of spontaneous activity in mice(P<0.05), and showed synergic effect on pentobarbital-induced sleeping by increased numbers of mice falling asleep(P<0.05), reduced the sleep latency and prolonged the sleeping time(P<0.01). Futhermore, Low doses of peanut red skin water extract increased the content of GABA in brain of mice(P<0.01). Conclusion The peanut red water extract has a function of sedative and hypnotic and one of its mechanisms may be to increase the content of GABA in brain of mice. |
