      目的 制备鱼藤酮微球, 并对其质量进行评价。方法 采用乳化-溶剂挥发法,制备鱼藤酮微球,利用扫描电镜观察其形态及粒度分布,HPLC法测定包封率、载药量、体外释药曲线。按2.5mg/kg剂量给予大鼠颈部皮下注射鱼藤酮微球,对体重变化和行为活动进行统计和观察。结果 所得的微球扫描电镜下呈圆球形,平均粒径9.58nm,载药量为40.39%,包封率为85.72%,体外释药可达7d,释药曲线符合一级动力学方程。微球给药方式对大鼠体重增长影响较小,给药后大鼠出现动作迟缓、弓背状姿势等帕金森病模型的典型特征。结论 本实验制备得到的鱼藤酮微球体外具有良好的缓释作用,微球给药显著降低了给药频率和造模死亡率,有望用于帕金森大鼠模型的研究。

      Abstract Objective To prepare rotenone microspheres and evaluate the quality. Methods Rotenone microspheres were prepared by emulsion-solvent evaporation method.The morphology and particle size distribution were observed by scanning electron microscope.The entrapment efficiency,drug loading rate and in vitro release rate were determined by HPLC.The rotenone microspheres were injected subcutaneously into the neck of rats at the dose of 2.5mg/kg, and the changes of body weight and behaviors were observed. Results The microspheres were spherical, with an average particle size of 9.58nm,drug loading rate of 40.39%,entrapment efficiency of 85.72%,and it could release for 7days in vitro. The release curve accorded with the first order kinetic equation. The microspheres drug delivery system had little effect on the growth of body weight in rats, and typical characteristics of Parkinson's disease were observed in rats. Conclusion Rotenone microspheres showed good sustained release effect in vitro, and it could significantly decrease the frequency of administration and the mortality of model rats, and was expected to be used in the establishment of Parkinson’s rat model.
