      目的 对日照海域野生牡蛎共生真菌进行分离、鉴定,通过活性筛选获得活性菌株。方法 从日照海域采集野生牡蛎,对其共生真菌进行分离、鉴定,构建系统发育树,采用DPPH清除自由基法和Ellman法研究其代谢产物的抗氧化和抑制乙酰胆碱酯酶(AChE)活性。结果 从牡蛎中共分离得到17株共生真菌,分别为青霉菌Penicillium sp.(Y-1),丝衣霉菌Byssochlamys sp.(Y-2、Y-6、Y-9、Y-13),链格孢菌Alternaria alternata(Y-3、Y-7);篮状菌属Talaromyces radicus(Y-4),黄曲霉Aspergillus flavus(Y-5),红曲霉Aspergillus ruber strain(Y-8、Y-11),芽枝孢霉菌Cladosporium perangustum strain(Y-10、Y-17),宛氏拟青霉Paecilomyces variotii strain(Y-12),绳状篮状菌属Talaromyces funiculosus strain(Y-14),聚多曲霉Aspergillus sydowii(Y-15),姬松茸,蘑菇科真菌Agaricaceae sp.(Y-16)。多数菌株的代谢产物具有一定的抗氧化活性和抑制AChE活性作用,其中,Y-8和Y-11的代谢产物的活性较为显著,浓度为200μg/ml时对DPPH自由基清除率分别达38.79%和48.27%,对AChE活性抑制率分别达39.20%和40.98%。结合HPLC分析,发现生物活性好的菌株其次级代谢产物的丰富度也较高。结论 本研究得到2株具有代谢产物丰富、生物活性显著的真菌,为后续具有抑制AChE活性和抗氧化活性的天然先导化合物的分离奠定基础。
英文摘要:
      Objective To isolate and identify endophytic fungi of oysters from Rizhao offshore area and the activities of antioxidant and AChE-inhibitory were determined.Methods Endophytic fungi from wild oysters were isolated and purified and the phylogenetic tree was constructed and discussed.Further,the antioxidant activity and inhibition of AChE activity were studied by DPPH free radical scavenging assay and Ellman's method respectively.Results 17 strains fungi had been isolated from wild oysters and belonged to eleven species of fungi that were Penicillium sp.(Y-1),Byssochlamys sp.(Y-2,Y-6,Y-9,Y-13),Alternaria alternate (Y-3,Y-7),Talaromyces radicus (Y-4),Aspergillus flavus (Y-5),Aspergillus ruber strain (Y-8,Y-11),Cladosporium perangustum strain (Y-10,Y-17),Paecilomyces variotii strain (Y-12),Talaromyces funiculosus strain (Y-14),Aspergillus sydowii (Y-15),Agaricaceae sp.(Y-16).Biological activities tests indicated most metabolites of strains showed antioxidation effect and the inhibition of AChE activity,with strains Y-8 and Y-11 having the significant activities.The scavenging rates of DPPH free radicals and inhibition rates of AChE activity were 38.79%,48.27% and 39.20%,40.98% at the concentration of 200μg/mL.Combined with HPLC analysis,it was found that the metabolites of the strains with good biological activity were also abundant.Conclusion In this study,two strains of fungi with abundant metabolites and remarkable biological activity were obtained.These bioactive strains provided much valuable leading compounds with antioxidant and AChE-inhibitory activity.
