      目的 探究IL-16在恶唑酮诱导的小鼠炎症性肠病(inflammatory bowel disease,IBD)模型中的表达及意义。方法 雌性7周龄C57BL/6小鼠随机分为正常对照组和模型组,每组10只。正常对照组做乙醇对照处理,模型组经恶唑酮诱导建立小鼠IBD模型。观测小鼠活动度以及便血情况并记录体重变化,7d后眼球静脉取血处死小鼠,分离小鼠结肠组织,制作结肠组织HE切片染色,q-PCR检测小鼠结肠组织中细胞因子IL-16 mRNA的变化,ELISA检测小鼠结肠组织中IL-16蛋白水平的变化。结果 与正常对照组相比,模型组小鼠结肠组织中IL-16 mRNA的水平明显升高,差异具有统计学意义(P<0.05);模型组IL-16蛋白水平明显升高,差异有统计学意义(P<0.05)。结论 IL-16在恶唑酮诱导的小鼠IBD模型中高表达,可能参与小鼠IBD的发生发展。
英文摘要:
      Objective To investigate the expression and significance of IL-16 in the model of oxazolone-induced inflammatory bowel disease in mice.Methods Twenty female 7-week-old C57BL/6 mice were randomly divided into normal control group and model group.The normal control group was treated with Ethanol control and the model group was induced by oxazolone to establish an inflammatory bowel disease model in mice.The mouse activity and blood in the stool were observed and body weight changes were recorded.Seven days later,the mice were sacrificed by intraocular vein blood sampling.Colon tissue was isolated from mice for HE section staining.q-PCR was used to detect the change of cytokine IL-16 mRNA in colon tissue,and ELISA was used to detect the changes of IL-16 protein in colon tissue of mice.Results Compared with the normal control group,the level of IL-16 mRNA in the colon tissue of mice in the experimental model group was significantly increased (P<0.05).The level of IL-16 protein in the model group was also increased and the difference was statistically significant (P<0.05).Conclusion IL-16 is highly expressed in oxazolone-induced inflammatory bowel disease in mice which is involved in the development of inflammatory bowel disease in mice.
