目的 通过检测自噬相关基因在大鼠脑缺血-再灌注损伤模型中的差异表达,探讨自噬在脑缺血-再灌注损伤过程中的作用及机制。方法 线栓法制备大鼠局灶性脑缺血-再灌注损伤模型;RNA-sequencing方法筛选大鼠脑缺血-再灌注24h模型中差异表达的自噬基因,并利用qRT-PCR方法进行验证;RT-PCR方法检测差异自噬基因在脑缺血2 h及不同再灌注时间点(6、12、24、48h)的表达变化。结果 TTC染色表明成功制备大鼠局灶性脑缺血-再灌注损伤模型;自噬相关基因在大鼠脑缺血-再灌注模型中差异表达,而且随着再灌注时间的延长其表达出现先升高后下降的趋势。结论 自噬相关基因参与了脑缺血-再灌注损伤,为进一步揭示自噬在脑缺血-再灌注损伤中的机制提供实验基础。 |

Objective To identify differentially expressed genes related to autophagy in a rat model of cerebral ischemia/reperfusion (I/R) injury and elaborate the molecular mechanism of autophagy in cerebral I/R injury.Methods Firstly,a rat model of focal cerebral I/R injury was established which has been ischemia for 2h and reperfusion for 24h.RNA-sequencing was used to identify differentially expressed genes and qRT-PCR was used to further detect the expression of different genes related to autophagy.RT-PCR was used to amplify genes related to autophagy at different times of reperfusion.Results Compared to the sham group,the infarct areas appeared white when stained with TTC in model groups.The genes related to autophagy were differentially expressed in rat I/R model.Furthermore,the expression patterns of different genes displayed significant differences at various times during the post-reperfusion period.Conclusion The results showed that autophagy may be involved in cerebral I/R injury,which provides an experimental basis for further revealing the mechanism of cerebral I/R injury. |
