      目的 优选复合酶法提取玄参多糖的最佳酶配比工艺条件,并对玄参多糖体外抗氧化活性进行初步研究。方法 以玄参多糖得率为考察指标,在单因素基础上,采用正交实验设计优化果胶酶、纤维素酶、半纤维素酶和木瓜蛋白酶的用量,并在最佳酶配比下,考察料液比、pH值、提取时间、酶作用温度的最佳提取工艺;利用1,1-二苯基-2-三硝基苯肼(DPPH·)法测定玄参多糖抗氧化活性。结果 复合酶最佳配比为:果胶酶1.50%、纤维素酶1.20%、木瓜蛋白酶0.90%、半纤维素酶0.09%,最佳提取条件为:料液比为1:30(g/ml)、pH值为7.0、反应时间为60min、温度40℃。在此条件下,玄参多糖含量达到8.98%。玄参多糖对DPPH·清除作用在0.5~2.5mg/ml浓度范围内呈较强的量效关系。结论 酶辅助提取方法显著提高了玄参多糖的提取率,可为玄参多糖的提取、分离奠定了良好的基础。

      Objective To explore the best compound enzymes ratio,the optimal extraction technology on the Scrophularia ningpoensis polysaccharides (SNP) from S.ningpoensis Hemsl.,and preliminary antioxidant activities of SNP.Methods Using extraction rate of polysaccharide as index,the orthogonal design based on single factor test was employed to further optimize the proportion of various enzymes (pectinase,cellulose,papain,hemicellulase) and the effects of extraction conditions (ratio of water to raw material,pH,extraction time,extraction temperature).In vitro the antioxidant activity of polysaccharide was determined by DPPH free radical scavenging method.Results The optimal amounts of complex enzymes were:pectinase of 1.50%,cellulose of 1.20%,papain of 0.90% and hemicellulase of 0.09%.The results showed that the highest polysaccharides yield of 8.98% was achieved with extraction time of 60 min,extraction temperature of 40℃,pH value of 7.0,liquid/material ratio of 30:1 ml/g.SNP using complex enzyme had significant antioxidant activity,and the DPPH radical-scavenging rate assumed a rise trend along with the increase of polysaccharide concentration in the range of 0.5-2.5mg/ml.Conclusion The findings showed that the complex enzyme-assisted extraction improved the yield of SHP and provided the basis for extraction and separation of SNP from Scrophularia ningpoensis.
