| 目的 建立HPLC-DAD方法，同时测定苦荞麦保健品中芦丁、槲皮素及山奈酚的含量。方法 采用色谱柱TOSOH TSK gel ODS-100V（4.6×150 mm，3.0 μm），以乙腈-水（含0.2%冰醋酸）为流动相，梯度洗脱，柱温35℃，流速1.0 ml/min，检测波长254 nm。结果 建立的HPLC-DAD分析方法，能够满足苦荞麦保健品中芦丁、槲皮素及山奈酚的完全分离，芦丁、槲皮素及山奈酚在10~500 μg/ml范围内有良好的线性关系r2均大于0.9997，重复性相对标准偏差（RSD）<2.3%，精密度RSD<1.5%，加样回收率为97.2%~102.8%，检出限和定量限分别为1μg/ml和5μg/ml。结论 该方法简便可行，测定结果准确，能够满足苦荞麦保健品中主要黄酮类化学成分测定的需要。 |

| Objective To establish a high performance liquid chromatography with a diode array detector (HPLC-DAD) method for simultaneous determination of rutin,quercetin,and kaempferol in tartary buckwheat functional food.Methods The chromatographic separation was performed on a TOSOH TSK gel ODS-100V (4.6×150mm,3.0μm) column.The mobile phase was composed of water containing 0.2% acetic acid (A) and acetonitrile (B) using gradient elution.The column temperature was maintained at 35℃;the flow rate was set at 1.0 ml/min;and UV absorbance was detected at a wavelength of 254 nm.Results The HPLC-DAD method was established and validated to determine the rutin,quercetin,and kaempferol in tartary buckwheat functional food.The calibration curves of rutin,quercetin,and kaempferol were in the range of 10~500μg/ml,with good linearity (r2>0.9997),repeatability (RSD<2.3%),precision (RSD<1.5%),recovery (97.2%~102.8%),limit of detection (1μg/ml),and limit of quantification (5μg/ml).Conclusion The method is a simple,feasible,and accurate approach for the determination major flavonoids in tartary buckwheat functional food. |
