      目的 探讨羟苯磺酸钙在慢性肾衰模型大鼠肾脏纤维化进程中的抑制作用。方法 将22只SD大鼠随机分为假手术组(n=7)、模型组(n=8)和治疗组(n=7)。采用5/6肾切除术建立慢性肾衰竭模型, 4周后假手术组和模型组给予饮用水, 治疗组给予羟苯磺酸钙灌胃, 监测各组大鼠血肌酐(serum creatinine, Scr)、尿素氮(urea nitrogen, BUN)、尿酸(UA)、C反应蛋白(C-reative protein, CRP)、胱抑素C(cystatin C, CysC)、透明质酸(uric acid, HA)、层粘连蛋白(laminin, LN)、Ⅲ型前胶原(procollagen-Ⅲ, PC-Ⅲ)、Ⅳ型胶原(collagen-Ⅳ, C-Ⅳ)等指标, 并观察肾组织中趋化因子受体CXCR4的表达情况。结果 与模型组比较, 羟苯磺酸钙治疗组大鼠Scr、BUN、HA、LN、PC-Ⅲ、C-Ⅳ等指标均明显减少, 差异有统计学意义;模型组大鼠肾组织中CXCR4免疫组化IOD值(125.43±5.35), 较假手术组增多, 差异有统计学意义;治疗组肾组织中CXCR4免疫组化IOD值为(82±3.26), 较模型组减少, 差异有统计学意义。结论 羟苯磺酸钙可延缓肾脏纤维化进程, CXCR4可能参与肾纤维化的过程。
英文摘要:
      Objective To study the inhibition of calcium dobesilate on renal fibrosis in a rat model of renal failure.Methods 22 SD rats were randomly divided into sham-operated groups (n=7), model groups (n=8) and treatment groups (n=7).Built by 5/6 nephrectomy renal failure model, the sham-operated group and model group were given drinking water after the surgery for 4 weeks, and the treatment group was given calcium dobesilate IG.In rat serum we measured the following:creatinine (Scr), urea nirtogen(BUN), uric acid(UA), C-reactive protein(CRP), cystatin C(CysC), hyaluronic acid (HA), laminin (LN), typeⅢ procollagen peptide (PC-III), typeⅣcollagen (C-IV) and CXCR4 expression in renal tissue.Results Compared with sham-operated groups, Scr、BUN、HA、LN、PC-Ⅲ、C-Ⅳand other indicators of the treatment group were significantly reduced, and the difference was statistically significant.In rat renal tissues of model group immunohistochemical IOD value of CXCR4 was 125.43±5.35 which is higher than sham-operated group, and the difference was significant.In the treatment group IOD values of CXCR4 in renal tissues CXCR4 by immunohistochemistry was 82± 3.26 which was lower than model Group, and the difference was significant.Conclusion Calcium dobesilate can delay the process of renal fibrosis, and CXCR4 may be involved in the process of renal fibrosis.
