| 目的 研究枸骨叶中芦丁的最佳提取工艺,并建立其含量测定方法。方法 以芦丁含量为指标,采用L9(34)正交设计,考察了提取时间、乙醇浓度、料液比、提取次数对提取率的影响,优选出合理的提取工艺。用高效液相法测定芦丁的含量。结果 最佳提取工艺条件为:用10倍量的90%乙醇,超声提取2次,每次50 min,芦丁在10.70~64.20 μg/ml范围内具有良好的线性关系(r=0.9999),平均加样回收率为98.70%,RSD为2.43%。结论 该提取工艺简便、快捷,合理可行,测定方法准确,重现性好,专属性强,为枸骨叶质量标准的建立提供了科学依据。 |

| Objective To study the extraction process and determination of rutin in the leaves of Ilex cornuta.Methods With rutin as index,extraction time,concentration of alcohol,amount of alcohol and extraction times were evaluated by L9(34) orthogonal design method.HPLC was used to analyze the content of rutin.Results The best extraction process was to extract 2 times with ten times of 90% ethanol for 50 minutes each time.Rutin showed a good linear relationship at the range of 10.70~64.20μg/ml(r=0.9999).The recovery rate was 98.70% and RSD was 2.43%.Conclusion The extraction method is simple,rapid and applicable.The HPLC determination was shown to be rapid,reliable and simple,and may be used for the quality control of the leaves from Ilex cornuta. |
