| 目的 检测胃癌及癌旁组织中Wnt10b的表达,探讨其与临床病理参数的关系。方法 收集25例胃癌患者手术切除的癌组织及对应的癌旁组织,用实时荧光定量PCR检测 Wnt10b mRNA的表达,Western-blot技术检测Wnt10b蛋白的表达,分析Wnt10b mRNA在胃癌组织中的表达与临床病理参数的关系。结果 胃癌组织中Wnt10b mRNA和蛋白表达水平明显高于癌旁组织(P<0.001),胃癌组织中Wnt10b mRNA表达阳性率与肿瘤远处转移相关(P<0.05),但与年龄、性别、肿瘤大小、TNM分期、分化程度和浸润深度无关。结论 Wnt10b在胃癌组织中呈活化状态,可能参与胃癌的发生发展。 |

| Objective This study aimed to detect the expression of Wnt10b in paired tumor and non-cancerous tissues of gastric cancer(GC) patients and explore correlation of Wnt10b mRNA expression with the clinicopathological parameters of GC.Methods The surgically resected paired cancerous and adjacent non-cancerous tissues of 25 GC patients were collected.The expression of Wnt10b mRNA was examined by Quantitative real-time PCR(qRT-PCR).Western-blot was employed to detect Wnt10b protein levels.Correlation between Wnt10b mRNA expression and clinicopathological parameters was analyzed.Results The levels of Wnt10b mRNA in cancerous tissues were higher compared with matching adjacent non-cancerous tissues(P<0.001).The protein levels of Wnt10b were consistent with the expression levels of Wnt10b mRNA.Wnt10b mRNA expression was correlated with the lymph node metastasis of tumor(P<0.05),but no association was found between Wnt10b mRNA expression and other clinicopathological parameters including age,gender,tumor size,TNM stage,differentitation stage and depth of invasion.Conclusion The expression levels of Wnt10b in cancerous tissues were higher than adjacent non-cancerous tissues,and there was correlation between the expression levels of Wnt10b and the lymph node metastasis of tumor. |
