      目的 探究芹菜素对人食管癌Eca-109细胞增殖及侵袭的影响。方法 体外培养食管癌Eca-109细胞,随机分为芹菜素低、中、高组和空白对照组,MTT法检测芹菜素对常规培养的食管癌Eca-109细胞的生长抑制情况;流式细胞仪Annexin V/PI双染色法检测细胞凋亡以及对细胞周期的影响;Transwell侵袭实验观察芹菜素对Eca-109细胞侵袭、运动能力的影响。结果 MTT法结果显示,各芹菜素组对Eca-109细胞的生长抑制作用随剂量增加逐渐增强,呈现剂量依赖性,在24、48、72h,低剂量组的抑制率分别为11.28%、19.45%、37.78%,中剂量组的抑制率分别为14.47%、29.49%、49.8%,高剂量组的抑制率分别为15.78%、31.21%、67.21%,对照组在各时间段无抑制情况,实验组与对照组比较差别具有统计学意义(P<0.001)。Transwell侵袭实验显示,48h后芹菜素低、中、高剂量组和空白对照组细胞穿膜数分别为(78.50±3.48)个、(55.50±4.41)个、(37.75±3.36)个和(90.75±5.32)个,各芹菜素组与空白对照组比较差别具有统计学意义(P<0.001),不同浓度芹菜素组间两两比较具有统计学意义(P<0.001)。结论 芹菜素对人食管癌109细胞生长有抑制作用,能明显抑制其侵袭和运动能力。

      Objective To investigate the effect of apigenin on proliferation and invasion of human esophageal cancer Eca-109 cells in vitro.Methods In vitro culture of Eca-109 cells from esophageal cancer,logarithmic cells were taken for experiments.The inhibitory effect of apigenin on the growth of Eca-109 cells cultured in vitro by MTT assay,FlowcytometryAnnexin V/PI double staining for cell apoptosis and its effect on cell cycle,Transwell Invasion Test to Observe the Effect of Apigenin on Invasion and Motility of Eca-109 Cells.Results In MTT results,the experimental groups(5mg·L-1、10mg·L-1、20mg·L-1) were treated with apigenin,inhibitory effect on Eca-109 cell growth gradually increased in a dose-dependent manner.The inhibition rates of low dose group were 11.28%,19.45% and 37.78% at 24,48 and 72 hours respectively,the inhibition rates of middle dose group were 14.47%,29.49% and 49.8%at 24,48 and 72 hours respectively.The inhibition rates of high dose group were 15.78%,31.21% and 67.21% at 24,48 and 72 hours respectively in a time-dependent manner.There was no inhibition in the control group at all time intervals.The difference between the experimental group and the control group was statistically significant(P<0.001).There was statistical significance between two groups of different concentration and different time(P<0.001).Transwell Invasion Experiments showed that the number of cell membrane penetration in the control group was 90.75±5.32.The number of cell membrane penetration in low dose group was 78.50±3.48.The number of cell membrane penetration in the medium dose group was 55.50±4.41.The number of cell membrane penetration in the high dose group was 37.75±3.36.The difference between the experimental group and the control group was statistically significant(P<0.001).There was statistical significance between two groups with different concentrations(P<0.001).Conclusion Apigenin can inhibit the growth of human esophageal cancer 109 cells and significantly inhibit its invasion and motor ability.
